In weak phase case, the specimen is much thinner than the mean free path for all the inelastic scattering processes. Many biological TEM specimens practically satisfy this criterion at thicknesses < 50 nm, but the specimens containing heavier atoms need to be much thinner than 50 nm. In general, thin biological, frozen-hydrated specimens can be modeled as weak phase objects.
